IGF-I and IGF-II mRNA expression in primary cultures of fetal hepatocytes

Abstract

Secretion of IGFs in adult animals is mainly regulated by growth hormone (GH),

but the role of other factors becomes principal during the perinatal period of the rat,

when the IGF response to GH is not yet fully established. In vitro models overcome

the simultaneous fluctuations of fuels and hormones that occur in live animal s and are

useful to investigate the underlying mechanisms by which hormones and nutrients

regulate IGF secretion. A selective primary culture of fetal rat hepatocytes has been

established in our laboratory in order to elucidate the molecular mechanism of action

of different factors and conditions on IGF-I and -11 gene expression during the perinatal

period of the rato In this preliminary study we report the regulatory role of semm on

IGF-I and -11 gene expression during stages of perinatal development. This system

should be a useful tool for further studies of molecular mechanisms of IGF-I and -11

regulation.
