Formulation of unidirectional Release Buccal Patches of Carbamazepine and study of permeation through porcine buccal mucosa.

Resumen

Objetivo: El objetivo del presente trabajo es estudiar la liberación de carbamazepina transbucaldesde parches bucoadhesivos.Métodos: Se preparan parches bucales de carbamazepina utilizando HPMC K-15M, PVA, PVP K-30y EC por el método de dispersión de solventes. Se estudiaron seis formulaciones (FA2, FA8, FA10,FB1, FB14 y FB16), como capa de soporte impermeable de parches, se utiliza Pidilite ® BOPP film.Se evaluaron el espesor, uniformidad de masa, pH superficial, resistencia al plegado. Se determinóla inflamación, resistencia mucoadhesiva, tiempo de mucoadhesión, liberación y permeación delfármaco así como estudios de estabilidad acelerada.Resultados: El tiempo de mucoadhesión osciló entre los 109 min (FA10) y los 126 min (FB14). Laresistencia mucoadhesiva está en el intervalo de 0,278 a 0,479 Kg.ms-2. Los estudios in vitro de liberaciónde fármaco mostraron que la formulación FA8 libera 84% y la FB16 libera 99,01% en 140 min.Conclusiones: Los parches bucales de carbamazepina diseñados, cumplen con los requisitos demucoadhesión adecuados para poder considerar este sistema como alternativa en la administraciónde carbamacepina.
